INVESTIGATOR REQUIREMENTS FOR BLASTOCYST INJECTIONS
* A completed application
form and copy of your current ACUC approval letter must be submitted prior
scheduling injections
* Evidence that your parental ES cell line had
been tested (MAP, Mouse Antibody Production or newer PCR-based assay) for
mouse pathogens that may infect our mouse colony
* Mycoplasma results on individual clones
* It is also recommended that clones be analyzed
for chromosomal content
* ES cells prepared for injection outside the facility must be observed while in culture (prior to trypsinization) by TTML staff on the day of injection so that morphology can be evaluated
BLASTOCYST (CHIMERIC MOUSE PRODUCTION) SERVICES PROVIDED BY THE LABORATORY
* Chimeric mice are generated by injecting targeted ES cell clones into E3.5 C57BL/6 blastocysts and transferred blastocysts into 2.5d recipient females. We guarantee injection of 50 blastocysts per clone and will provide any resulting chimeric pups
* ES cells prepared for injection outside the facility must be observed while in culture (prior to trypsinization) by TTML staff on the day of injection so that morphology can be evaluated
* Germline transmission cannot be guaranteed for ES cells manipulated outside the Laboratory
